Effetti dell’esposizione precoce alla dieta occidentale e dell’esercizio volontario sui livelli di attività degli adulti, sulla fisiologia dell’esercizio e sui tratti associati nei topi High Runner selezionati in modo selettivo

Il comportamento durante l’esercizio è sotto il controllo genetico parziale, ma è anche influenzato da numerosi fattori ambientali, potenzialmente comprese le prime esperienze di vita i cui effetti persistono nell’età adulta. Abbiamo studiato gli effetti ambientali genetici e nei primi anni di vita sul comportamento della ruota in un modello murino che embody quattro linee di excessive runner (HR) replicate selettivamente allevate per aumentare la corsa su ruota volontaria da giovani adulti e quattro linee di controllo (C) non selezionate. In un disegno fattoriale completo, ai topi di ciascuna linea è stato concesso o meno l’accesso alla ruota e sono stati somministrati dieta commonplace o occidentale (WD) dallo svezzamento (three settimane di età) a 6 settimane di età (maturità sessuale).

Oltre agli effetti acuti, dopo un periodo di washout di eight settimane (∼6 anni umani) in cui tutti i topi avevano una dieta commonplace e nessun accesso alla ruota, abbiamo riscontrato effetti sia benefici che dannosi di queste esposizioni precoci. Durante la prima settimana di trattamento, la WD ha aumentato la distanza percorsa del 29% nei topi C e del 48% nei topi HR (significativa interazione dieta × tipo di linea), ma gli effetti della dieta sono scomparsi entro la terza settimana. Durante le tre settimane di trattamento giovanile, la WD ha aumentato significativamente la massa grassa (con la massa magra come covariata).

Testato da adulti, l’esercizio fisico nella prima infanzia ha aumentato la corsa delle ruote dei topi C ma non dei topi HR nella prima settimana. L’esercizio fisico nella prima infanzia ha anche ridotto il comportamento ansioso degli adulti e aumentato i livelli di glucosio nel sangue a digiuno degli adulti, la massa del tricipite surale, la massa del cuscinetto adiposo sottocutaneo e la massa cerebrale, ma ha ridotto la massa del ventricolo cardiaco. Utilizzando la massa grassa come covariata, il trattamento con esercizi fisici nei primi anni di vita ha aumentato la concentrazione di leptina negli adulti. Al contrario, la WD in età precoce ha aumentato la corsa della ruota degli adulti dei topi HR ma non dei topi C. La WD in età precoce ha anche aumentato la massa magra degli adulti e la preferenza degli adulti per la dieta occidentale in tutti i gruppi.

Sorprendentemente, il trattamento nella prima infanzia non ha avuto effetti significativi sul grasso corporeo adulto o sulla capacità aerobica massima (VO2max). Nessuno studio precedente ha testato gli effetti combinati o interattivi della WD e dell’esercizio fisico nella prima infanzia. I nostri risultati dimostrano che entrambi i fattori possono avere effetti di lunga durata sull’esercizio volontario degli adulti e sui relativi fenotipi e che questi effetti sono modulati dal background genetico. Nel complesso, gli effetti a lungo termine dell’esercizio fisico precoce erano più pervasivi di quelli della WD, suggerendo opportunità critiche per l’intervento sanitario nelle abitudini infantili, nonché possibili minacce derivanti dalle sfide moderne. Questi risultati possono essere rilevanti per comprendere i potenziali effetti della riduzione dell’attività e dei cambiamenti nella dieta associati all’epidemia di obesità e alla pandemia di COVID-19.

Impatti dei cambiamenti nell’O 2 atmosferico sulla fisiologia umana. C’è un motivo di preoccupazione?

Viene spesso espressa preoccupazione per la continua perdita di O2 atmosferico. È probabile che questa perdita, causata dalla combustione di combustibili fossili ma anche influenzata da altri processi, continui almeno per i prossimi secoli. Sosteniamo che questa perdita è abbastanza ben compresa e l’eventuale diminuzione è limitata dalla base delle risorse di combustibili fossili. Poiché il serbatoio di O2 atmosferico è così grande, il calo relativo previsto di O2 è molto piccolo anche per scenari estremi di utilizzo futuro di combustibili fossili che producono aumenti della CO2 atmosferica sufficienti a causare cambiamenti climatici catastrofici.

A livello del mare, la caduta finale della pressione parziale dell’ossigeno sarà inferiore a 2,5 mm Hg su una linea di base di 159 mmHg. È probabile che il calo entro il 2300 sia compreso tra 0,5 e 1,three mmHg. Le implicazioni per la normale salute umana sono trascurabili perché il consumo di O2 respiratorio in individui sani dipende solo debolmente dalla pressione parziale ambientale, specialmente a livello del mare. Anche gli impatti sulle prestazioni dei migliori atleti, sulla malattia, sulla riproduzione e sulla cognizione saranno molto piccoli. Fistole arteriose coronariche acquisite e progressive in pazienti con fisiologia a ventricolo singolo e trattati con vasodilatatori polmonari

Disfunzione cardiaca, aritmia e fibrosi epatica sono complicazioni ben observe dopo l’intervento chirurgico di bypass del cuore destro in pazienti con fisiologia a ventricolo singolo. Tuttavia, si sa poco sulle fistole arteriose coronariche e sono stati pubblicati solo pochi rapporti. Questo studio mirava a chiarire le caratteristiche cliniche di queste uncommon fistole arteriose coronariche che si sono sviluppate come complicanze nei casi di fisiologia del ventricolo singolo dopo un intervento di bypass cardiaco destro.

abbiamo studiato retrospettivamente le caratteristiche cliniche e il decorso dei pazienti che hanno sviluppato fistole arteriose coronariche acquisite e progressive dopo un intervento di bypass del cuore destro nel nostro ospedale. abbiamo identificato tre casi di fistole arteriose coronariche su 21 pazienti sottoposti a intervento chirurgico di bypass del cuore destro. Tutti e tre i casi sono stati sottoposti a cateterizzazione cardiaca per la valutazione post-operatoria e sono stati somministrati vasodilatatori polmonari di inibitori della fosfodiesterasi di tipo V, antiaggreganti piastrinici, anticoagulanti e diuretici. Analisi comparativa di morfologia, fisiologia fotosintetica e trascrittoma tra orzo diploide e tetraploide derivato da coltura microspore

I poliploidi svolgono un ruolo importante nell’allevamento di piante per caratteristiche superiori e molti rapporti si sono concentrati recentemente sugli effetti sulla fotosintesi della poliploidizzazione in alcune specie vegetali, ma sorprendentemente poco di questo è noto per l’orzo. In questo studio, piante omozigoti diploidi e tetraploidi, derivate dalla coltura microspore della cultivar d’orzo “H30”, sono state utilizzate per valutare le differenze tra loro nelle loro caratteristiche cellulari, fotosintetiche e trascrittomiche. I nostri risultati hanno mostrato che l’orzo tetraploide ha le caratteristiche distintive dei poliploidi, vale a dire foglie più spesse e più pesanti,

dimensioni degli stomi ingranditi o dimensioni delle cellule di guardia stomatiche e più pigmenti fotosintetici e fotosintesi migliorata (specialmente in condizioni di elevata intensità di luce). Questa migliore fotosintesi dell’orzo tetraploide è stata confermata da diversi parametri fotosintetici, tra cui velocità fotosintetica netta (Pn), conduttanza stomatica (Gs), concentrazione intercellulare di CO2 (Ci), velocità di traspirazione (Tr), velocità fotosintetica netta massima (Pmax), saturazione della luce punto (LSP), carbossilazione a velocità saturata RuBP massima (Vcmax) e velocità massima di trasporto di elettroni (Jmax).

Analisi trascrittomiche hanno rivelato che solo il 2,3% circa di tutti i geni rilevati mostrava modelli di espressione differenziale [cioè geni espressi differenzialmente (DEG)], e che la maggior parte di questi – 580 su 793 DEG in totale – erano sovraregolati nell’orzo tetraploide.
